Abstract

This paper is an attempt to use a direct approach in the study of ultraviolet irradiation induced modifications of the covalent structure of bovine plasma albumin. The reactions to be considered will be largely those of the disulfide bonds because of their important role in recent theories on the structure of proteins. The fission of disulfide groups during ultraviolet irradiation is followed by amperometric titration of sulfhydryl groups formed in the fission process with mercuric chloride, silver nitrate, and ethyl mercuric chloride as titrants. These changes are then compared with changes of viscosity and optical rotation of albumin solutions upon irradiation.
