Abstract

It has been shown previously that HeLa cell nuclear extract can stimulate RNA/RNA annealing efficiently. The work presented here extended the study to the fractionation and characterization of the annealing activity. My results indicate that the activity is contributed by the protein component in the extract. It is also associated with a well-identified hnRNP (heterogeneous ribonuclear particle) protein Al. Similar to the crude extract annealing reaction, Al annealing operates under a wide range of salt, pH and temperature condition and has no apparent substrate specificity. Annealing is optimized at AJ/RNA binding saturation. Annealing reactions with various peptides of Al suggest that the Al annealing activity is mainly associated with the C-terminal domain. The N-terminal domain peptide UPl only displays very weak activity.
