Abstract
This work describes the construction and characterization of nine recombinant DNAs that contain sequences which are complementary to the early region 2A (E2A) transcript of adenovirus type 2 (Ad2). These clones are complementary to both intron and exon segments of this transcript and map between 63.6 map units (mu) and 70.2 mu on the Ad2 genome. These clones were constructed by ligating selected restriction fragments from this region to either the bacteriophage cloning vector Ml3mp8 or the plasmid vector pUC8. These clones make it possible to analyze the structure of the RNA transcribed from this region using high resolution methodologies that require small hybridization probes. By integrating the information obtained from such studies it should be possible to construct an accurate description of the structure of these sequences and to evaluate the potential of their structure to be of functional significance to the splicing mechanism.
